Trimethoprim inhibits the production of tetrahydrofolic acid, an essential component for the synthesis of DNA, RNA, and proteins in bacteria. By blocking this pathway, trimethoprim disrupts bacterial metabolism and prevents their ability to multiply and survive.
Sulfamethoxazole, on the other hand, is a sulfonamide antibiotic. It acts by inhibiting the production of dihydrofolic acid, another essential component required for bacterial growth. Sulfamethoxazole competitively binds to the enzyme responsible for synthesizing dihydrofolic acid, interfering with its function and ultimately inhibiting bacterial replication.
As a combination antibiotic, TMP-SMX provides a broad spectrum of coverage against various bacteria, making it effective against respiratory tract infections, urinary tract infections, gastrointestinal infections, skin and soft tissue infections, and certain sexually transmitted infections.
It is worth mentioning that TMP-SMX is commonly used as a first-line treatment for uncomplicated urinary tract infections caused by susceptible bacteria. However, resistance to this antibiotic may occur, so it is essential to conduct susceptibility testing before prescribing TMP-SMX. Additionally, TMP-SMX should be used with caution in individuals with known allergies to sulfonamide medications. Common side effects may include allergic reactions, gastrointestinal distress, and skin rash.
