DCFDA is an acronym that stands for Dichloro-dihydro-fluorescein diacetate. It refers to a chemical compound widely utilized in biological research to measure and assess cellular oxidative stress levels.
DCFDA is a non-fluorescent compound belonging to the class of fluorescein derivatives. It is typically present as a pale yellow solid consisting of tiny crystals. Once inside the cell, DCFDA is easily hydrolyzed by esterases, which remove the acetate groups to yield DCFH (Dichloro-dihydro-fluorescein).
DCFH is then oxidized by various reactive oxygen species (ROS), including hydrogen peroxide and peroxynitrite, within the cell, leading to the production of a highly fluorescent compound called DCF (Dichlorofluorescein). The intensity of fluorescence emitted by DCF is directly proportional to the amount of ROS generated in the cell, enabling researchers to evaluate the oxidative stress conditions.
